It works if you have it check marked in the Options Menu.

 

EDIT: Once checked, you can use it while in A2A modes as long as the aircraft you want to padlock is 1) within visual range and 2) more or less centered within your field of view. I don't know if it's still the case now but, in earlier iterations of the sim, you used to be able to padlock a point on the ground while in A2G mode as well.
