Is it possible to assign a pb"xx" key of a DDI on a hotas command (when there is no keyboard shortcut) ?

Yes it is, dunno which plane you are using,

 

For example, FA-18C

 

Go to controlls, the above tabs, first one is F/A-18C Sim second one says All, click on it, and scroll to

 

AMPCD = middle bottom screen

Left MDI = left screen

Right MDI = right screen

 

There you can fint the push buttons
